  2. Malwarebytes, Avast and Windows Defender

    Malwarebytes Anti-Malware (free version) is not an Anti-Virus product, see
    Does Malwarebytes Anti-Malware replace antivirus software?

    Therefore it will not interfere with whichever Anti-Virus product you want to use.

    But: You should not use more than 1 Anti-Virus product at the same time.

    The in Windows 10 in-built Anti-Virus program Windows Defender should be automatically disabled, as soon as any other Anti-Virus program is properly installed.

    You say you use Avast Premier and that Windows Defender "seems to be in a locked position of ON".

    That's not how it should be. Windows Defender should be off.

    I suggest to uninstall Avast with the help of their specific removal tool (https://www.avast.com/uninstall-utility), then reboot.

    And then re-install your Avast (make sure though beforehand that it's the latest Windows 10 compatible version) and reboot again.

    Now Windows Defender should be disabled.

    No, it does not release the malware. Be aware it will delete the quarantine and if there was a false positive, you would not be able to restore the files even if you reinstall MB.
    Second. Since Malwarebytes is now an antivirus "replacement" it will disable Defender like any other AV program if using the trial or the paid version.
    The trial is installed by default and if you are not going to use your key or purchase a subscription you need to deactivate the trial and defender will come back on.
    Or you can turn off Malwarebytes from registering in the Windows security center.
